{"id":5939,"date":"2018-05-21T20:51:30","date_gmt":"2018-05-21T18:51:30","guid":{"rendered":"http:\/\/agora.xtec.cat\/ies-merce-rodoreda\/?p=5939"},"modified":"2018-05-21T20:51:30","modified_gmt":"2018-05-21T18:51:30","slug":"icm-institut-de-ciencies-del-mar-del-csic","status":"publish","type":"post","link":"https:\/\/agora.xtec.cat\/ies-merce-rodoreda\/modul-a\/icm-institut-de-ciencies-del-mar-del-csic\/","title":{"rendered":"ICM &#8211; Institut de Ci\u00e8ncies del Mar del CSIC"},"content":{"rendered":"<p><iframe loading=\"lazy\" src=\"\/\/www.youtube.com\/embed\/0Yy6YDngpKg\" width=\"560\" height=\"314\"><\/iframe><\/p>\n<p>Nosaltres, la Sara i jo, hem realitzat les FCT a l&#8217;ICM (Institut de Ci\u00e8ncies del Mar) ajudant a la C\u00e8lia Marrass\u00e9, fent <strong>espectrofotometries<\/strong> i <strong>espectrofluorimetries<\/strong> de mostres d&#8217;aigua de mar, generalment.<\/p>\n<p>Duem a terme una s\u00e8rie d&#8217;experiments, pels que analitzem el <strong>FDOM<\/strong> (Fluorescent Dissolved Organic Matter) i el <strong>CDOM<\/strong> (Colored Dissolved Organic Matter).<\/p>\n<p>Per a explicar-vos el que fem a les FCT i com \u00e9s el nostre dia a dia a l&#8217;ICM, hem preparat un video i aix\u00ed pugueu veure que fem realment.<\/p>\n<p>&nbsp;<\/p>\n<p><!--more--><\/p>\n<p><span style=\"text-decoration: underline;\"><strong>PREPARACI\u00d3<\/strong><\/span><\/p>\n<p>Abans de cada an\u00e0lisi, s&#8217;ha de fer una preparaci\u00f3 de tot el material que ser\u00e0 utilitzat: les ampolles de 500 mL i de 250 mL de vidre on es posar\u00e0 la mostra, els pots de 30 mL per al TOC i DOC, la rampa de filtratge i les cubetes. Tot aquest material ser\u00e0 netejat 3 vegades amb aigua Milli-Q (MQ), que \u00e9s aigua pura; despr\u00e9s se li passa\u00a0HCl\u00a0al 1% i s&#8217;esbandeix una altra vegada 3 vegades amb aigua MQ.<\/p>\n<p>Tamb\u00e9 s&#8217;ha de dur a terme un etiquetatge apropiat, que consta del nom de l&#8217;experiment (DEAT, CAB, Deep-Long, VARITEC&#8230;), la data de l&#8217;experiment, qu\u00e8 es el que s&#8217;analitza (TOC, DOC, POC, POP&#8230;) i el n\u00famero de r\u00e8plica (I o II).<\/p>\n<p>Primer,\u00a0filtrem\u00a0la mostra en una rampa de filtratge, per gravetat i de vegades amb l&#8217;ajuda de nitrogen (que \u00e9s un gas inert), que aplica pressi\u00f3 sobre l&#8217;aigua per augmentar la velocitat de la filtraci\u00f3 i, posteriorment, analitzem el FDOM i CDOM de la mostra filtrada. Tamb\u00e9 es realitza un blanc de la mostra filtrada, \u00e9s a dir, es filtra aigua MQ que tamb\u00e9 s&#8217;analitzar\u00e0.<\/p>\n<p>A part, preparem 30 mL de les mostres per analitzar el TOC (Total Organic Carbon) i el DOC (Dissolved Organic Carbon), per\u00f2 aix\u00f2 es porta a un altre centre on s&#8217;analitzar\u00e0.<\/p>\n<p><span style=\"text-decoration: underline;\"><strong>FDOM (Fluorescent Dissolved Organic Matter)<\/strong><\/span><\/p>\n<p>En les espectrofluorimetries, analitzem el FDOM de les mostres. La mostra d&#8217;aigua es posa dins d&#8217;una cubeta de 1 cm de cam\u00ed \u00f2ptic, i obtenim 4 pics a l&#8217;espectrofluor\u00edmetre que pertanyen als h\u00famics totals, a les pseudo-prote\u00efnes, h\u00famics marins i h\u00famics terrestres que es troben dissolts a les mostres d&#8217;aigua. Aquests valors s\u00f3n la quantitat de mat\u00e8ria org\u00e0nica que hi ha a la mostra, i per tant, tamb\u00e9 la contaminaci\u00f3 d&#8217;aquestes mostres.\u00a0Primer, s&#8217;analitzen els dos blans (MQ i MQ filtrada) despr\u00e9s les mostres totals i les mostres filtrades.<\/p>\n<p><span style=\"text-decoration: underline;\"><strong>CDOM (Colored Dissolved Organic Matter)<\/strong><\/span><\/p>\n<p>Al dia seg\u00fcent, sempre analitzem el CDOM de les mateixes mostres d&#8217;aigua. Aquesta es fa en un esprectrofot\u00f3metre de doble feix, amb unes cubetes de 10 cm de cam\u00ed \u00f2ptic. En aquest cas, hi ha una cubeta de refer\u00e8ncia amb aigua Milli-Q que actua com a blanc i es resta directament al resultat de la mostra. Tot i aix\u00ed, tamb\u00e9 realitzem dos blancs (MQ i MQ filtrada). Es segueix el mateix ordre que a les espectrofluorimetries, primer els blancs, despr\u00e9s les mostres i les mostres filtrades.<\/p>\n<p>El primer que es fa \u00e9s la l\u00ednia base amb les dues cubetes de MQ, que elimina tot el soroll de fons de l&#8217;espectrofot\u00f2metre. Despr\u00e9s nom\u00e9s canviem una de les cubetes, on es posen les mostres, i es deixa durant tot l&#8217;an\u00e0lisi la cubeta de refer\u00e8ncia al mateix lloc sense canviar tampoc el seu contingut.<\/p>\n<hr \/>\n<p>Finalment, es neteja tot el material utilitzat seguint el mateix protocol que a l&#8217;inici. Es renta tot 3 vegades amb aigua MQ i se li passa \u00e0cid clorh\u00eddric l&#8217;1%. Es pot deixar amb l&#8217;\u00e0cid fins la pr\u00f2xima vegada que s&#8217;utilitzin aquestes ampolles.<\/p>\n<p>Les cubetes i la rampa de filtratge es deixen en un bany d&#8217;\u00e0cid clorh\u00eddric a l&#8217;1%, i quan s&#8217;hagin de tornar a utilitzar ho esbandirem amb molta aigua MQ.<\/p>\n<p>&nbsp;<\/p>\n<p>Per acabar, nom\u00e9s afegir que hem trobat una manera que les pr\u00e0ctiques no se&#8217;ns facin mon\u00f2tones, sempre intentem buscar la part positiva de totes les coses.
