En el temps que he estat aqu\u00ed he participat en diversos experiments i he treballat amb diverses t\u00e8cniques d\u2019an\u00e0lisi.<\/p>\n<p>En concret vaig a explicar un d\u2019aquests projectes. CAM \u00e9s el nom de la Campanya en la qual analitzem aigua de les illes Medes. S\u2019agafen mostres a quatre profunditats diferents: 0.5m, 20m, 50m i 80m. I les mesures que es duen a terme, en el meu departament \u00e9s per una banda, la Mat\u00e8ria Org\u00e0nica Dissolta, i per altra, uns pics espec\u00edfics (T: H\u00famics terrestres, M: H\u00famics marins, P: Pseudo-prote\u00efnes, A: H\u00famics generals).<\/p>\n<p style=\"text-align: justify;\"><!--more--><\/p>\n<h3 style=\"text-align: justify;\"><\/h3>\n<h3 style=\"text-align: justify;\">PRE-EXPERIMENT<\/h3>\n<p>El primer pas \u00e9s el rentat i etiquetatge del material que farem servir. En concret utilitzarem ampolles de policarbonat (de 30mL) i ampolles de vidre (de 250mL).<\/p>\n<p>Per a les ampolles de policarbonat el protocol es: esbandir amb aigua MQ (mili-Q) tant l\u2019interior com el tap. Omplir el fons amb HCL al 0.1%, agitar i deixar reposar uns 15min perqu\u00e8 l\u2019\u00e0cid pugui actuar. A continuaci\u00f3, tornar a rentar tres cops amb MQ i tancar.<\/p>\n<p>Per a les ampolles de vidre en canvi, el protocol es tenir-les guardades amb \u00e0cid uns dies abans del seu \u00fas, per la qual cosa el que s\u2019ha de fer \u00e9s retirar-lo: s\u2019esbandeixen les ampolles i els taps tres cops amb MQ.<\/p>\n<p>Per \u00faltim, etiquetem totes les ampolles amb el nom de CAM, la profunditat corresponent amb la que la omplirem i el dia de realitzaci\u00f3 de les mesures.<\/p>\n<h3 style=\"text-align: justify;\">MESURES A REALITZAR<\/h3>\n<p>Un cop tenim l\u2019aigua al laboratori, omplim les ampolles de vidre netes que ja tenim etiquetades amb la seva profunditat. \u00c9s important remoure una mica els bidons per homogene\u00eftzar l\u2019aigua, i tamb\u00e9 ambientar (esbandir tres cops amb l\u2019aigua de mostra) les nostres ampolles abans d\u2019omplir-les. El seg\u00fcent pas es filtrar part d\u2019aquesta aigua, ja que al analitzar l\u2019aigua fem la comparaci\u00f3 de l\u2019aigua pura i la filtrada. Els instruments que farem servir son el FDOM i el CDOM:<\/span><\/li>\n<\/ul>\n<p style=\"text-align: justify;\"><strong><span style=\"font-size: small;\">FDOM (Fluorescent Dissolved Organic Matter)<\/span><\/strong><\/p>\n<p>L\u2019equip que utilitzem \u00e9s un espectrofluor\u00edmetre Perkin Elmer, i el material: una cubeta de quars de 10mm d\u2019amplada de referencia de VWR, una cubeta de quars de 10mm de llargada de Perkin Elmer i les ampolles de vidre amb les aig\u00fces de mostra.<\/p>\n<p><span lang=\"ca-ES\">Els par\u00e0metres que analitzem amb aquest instrument s\u00f3n 4 pics: <\/span><span lang=\"ca-ES\">T (H\u00famics terrestres), M (H\u00famics marins), P (Pseudo-prote\u00efnes), A (H\u00famics generals)<\/span><\/p>\n<p><span lang=\"ca-ES\">El procediment a seguir en aquesta mesura \u00e9s en primer lloc encendre l\u2019ordinador i l\u2019espectrofluor\u00edmetre i deixar escalfar la l\u00e0mpada entre 20-30 minuts. Configurem l\u2019ordinador on es guardaran les dades i creem la carpeta corresponent. Obrim el programa (FL WinLab), amb el que es realitzaran les lectures. A continuaci\u00f3 ens posem els guants de vinil, traiem la cubeta del bany d\u2019\u00e0cid on est\u00e0 guardada i la esbandim amb MQ per retirar tot l\u2019\u00e0cid. El primer que mesurem ser\u00e0 MQ i MQ filtrada (blancs) per despr\u00e9s a la extracci\u00f3 de dades restar-ho a les mostres reals. Per omplir la cubeta, ambientem tres cops i la omplim deixant mig cent\u00edmetre buit i abans de posar-la dins de l\u2019espectrofluor\u00edmetre, comprovem que no te bombolles, saquem amb paper molt b\u00e9 totes les parets. La cubeta (d\u20191x1cm) te un petit segell en una cara, procurem sempre posar-lo cap al mateix sentit. Repetim el proc\u00e9s amb totes les mostres, fent tres replicats per cada una i tenint en compte que cada cop que canviem de mostra hem d\u2019ambientar tres cops la cubeta. Un cop em acabat totes les lectures, guardem la cubeta al bany d\u2019\u00e0cid i apaguem l\u2019espectrofluor\u00edmetre. Configurar l\u2019ordinador on es guardaran les dades i crear la carpeta corresponent. En aquest cas el programa que fem servir \u00e9s el Cary WinUV. Abans de res, configurem els l\u00edmits de la lectura de 250nm a 700nm, ja que es el rang que ens interessa. A continuaci\u00f3 ens posem els guants de vinil, agafem les dues cubetes (d\u20191x10cm!!) del bany d\u2019\u00e0cid i les netegem b\u00e9 amb MQ. La primera d\u2019aquestes ens servir\u00e0 per fer la l\u00ednia base (soroll de l\u2019instrument sense cap mostra) i durant totes les mesures es quedar\u00e0 dins de l\u2019instrument. La segona la farem servir per anar posant les mostres, i el primer que mesurem ser\u00e0 la MQ i la MQ filtrada, per fer servir de blancs. Les lectures tamb\u00e9 es fan igual que el cas anterior: ambientem sempre tres cops la cubeta abans d\u2019omplir-la amb cada mostra, omplim deixant mig cent\u00edmetre, vigilem que no hi hagi bombolles, saquem b\u00e9 les parets i procurem posar la marca de la cubeta sempre al mateix costat. A mesura que fem mostres ens van sortint les seves gr\u00e0fiques corresponents. Un cop fetes totes, apaguem l\u2019espectrofot\u00f2metre, netegem les cubetes i les tornem a l\u2019\u00e0cid. La bossa es congela a -20\u00baC (en un futur es determinar\u00e0 es carboni org\u00e0nic total). Els DOC es fan exactament igual, per\u00f2 s\u2019omplen amb les mostres d\u2019aigua filtrades per 0.7micres (en un futur es determinar\u00e0 es carboni org\u00e0nic dissolt).<\/p>\n<p style=\"text-align: justify;\"><span style=\"font-size: small;\">POC I POP<\/span><\/p>\n<p>Fem servir potets de 2mL, de criovial, marca NALGENE.<\/p>\n<p>Els potets s\u2019etiqueten, ambienten i omplen amb les aig\u00fces corresponents. Un cop acabat, es congelen a -80\u00baC.
