S\u00f3n capa\u00e7os de formar espores i el seu metabolisme \u00e9s sulfit reductor, ja que l&#8217;acceptor final d&#8217;electrons \u00e9s l&#8217;i\u00f3 sulfit. Viuen en aig\u00fces dolces i en el s\u00f2l. Tamb\u00e9 els trobem a l\u2019intest\u00ed, per\u00f2 en menor proporci\u00f3. En formar espores s\u00f3n molt resistents, et pel qual s\u00f3n indicadors d\u2019una contaminaci\u00f3 tardana de l\u2019aigua. S\u00f3n anaerobis perqu\u00e8 acceptor final d\u2019electrons de la seva cadena respirat\u00f2ria no \u00e9s l\u2019O<sub>2<\/sub>, sin\u00f3 l\u2019i\u00f3 sulfit (SO<sub>3<\/sub><sup>2-<\/sup>), que es redueix a i\u00f3 sulfur (S<sup>2-<\/sup>). El medi cont\u00e9 sulfit de sodi (Na<sub>2<\/sub>SO<sub>3<\/sub>); per a proporcionar als bacteris del seu acceptor final d\u2019electrons. Citrat f\u00e8rric (C<sub>6<\/sub>H<sub>5+4<\/sub>yFexNyO<sub>7<\/sub>) que en contacte amb l\u2019\u00e0cid sulfh\u00eddric forma sulfur de ferro (FeS) que \u00e9s un precipitat de color negre. La polixina i la sulfadiazina s\u00f3n antibi\u00f2tics selectius de bacteris gram<sup>&#8211;<\/sup>, per tal d&#8217;assegurar el creixement de bacteris gram<sup>+<\/sup>. El color negre caracter\u00edstic els el proporciona l&#8217;anteriorment esmentat FeS.<\/p>\n<p><span style=\"text-decoration: underline;\"><span style=\"font-weight: 400; color: #ff6600; text-decoration: underline;\">Objectius<\/span><\/span><\/p>\n<p><span style=\"font-weight: 400;\">Determinar la pres\u00e8ncia de clostridis sulfit-reductors a la mostra problema, d\u2019aigua del clavegueram.<\/span><\/p>\n<p><span style=\"text-decoration: underline;\"><span style=\"font-weight: 400; color: #ff6600; text-decoration: underline;\">Material<\/span><\/span><\/p>\n<ul>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">Gradeta<\/span><\/li>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">4 tubs d\u2019assaig<\/span><\/li>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">Pipeta graduada est\u00e8ril<\/span><\/li>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">4 tubs est\u00e8rils<\/span><\/li>\n<li style=\"font-weight: 400;\">Bany termoregulable<\/li>\n<\/ul>\n<p><span style=\"text-decoration: underline;\"><span style=\"font-weight: 400; color: #ff6600; text-decoration: underline;\">Reactius<\/span><\/span><\/p>\n<ul>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">Ringer lactat<\/span><\/li>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">Agar SPS<\/span><\/li>\n<\/ul>\n<p><span style=\"text-decoration: underline;\"><span style=\"font-weight: 400; color: #ff6600; text-decoration: underline;\">Procediment<\/span><\/span><\/p>\n<p><span style=\"font-weight: 400;\">Preparem 4 tubs amb agar SPS. Ho possem 20 minuts a l\u2019autoclau. Es preparen 4 tubs est\u00e8rils amb la mostra amb les dilucions pertinents (amb Ringer). Dep\u00e8n de la concentraci\u00f3 esperada podem fer un banc de dilucions mes concentrat o menys concentrat, en el nostre cas, com que esperem una alta concentraci\u00f3 (per la pr\u00f2pia naturalesa de la mostra) fem un banc de dilucions 1\/10, 1\/100 i 1\/1000. Per fer-ho introdu\u00efm 9 mL de Ringer en cadascun dels tres tubs d&#8217;assaig i en el primer (1\/10) afegim amb micropipeta 1 mL, d&#8217;aquest primer tub n&#8217;agafam 1 mL amb pipeta i el transvasem al segon i d&#8217;aquest \u00a0segon fem el mateix cap al tercer.\u00a0<\/span><span style=\"font-weight: 400;\"> Col\u00b7loquem els tubs en el bany termo-regulable a 80\u00baC durant uns 5-10 minuts.<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Inoculem amb la pipeta graduada est\u00e8ril 5 mL de la mostra de les anteriors dilucions i realitzem una inoculaci\u00f3 anaerobia. Es tracta d\u2019introduir la pipeta fins al fons del tub d\u2019assaig i anar traient mostra a mesura que es va pujant la pipeta (s\u2019ha de fer r\u00e0pid abans que solidifiqui l\u2019agar). Un cop s\u2019ha solidificat l\u2019agar, afegim 5 cm m\u00e9s d\u2019agar SPS o una capa de vaselina per a realitzar la capa d\u2019anaerobiosi.<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Ja podem posar els tubs a incubar a 37\u00baC durant 24 hores.<\/span><\/p>\n<p><span style=\"text-decoration: underline;\"><span style=\"font-weight: 400; color: #ff6600; text-decoration: underline;\">Observacions i Resultats<\/span><\/span><\/p>\n<p><span style=\"font-weight: 400;\">Si observem col\u00f2nies de color negre \u00e9s un indicador de la presencia de clostridis, voldr\u00e0 dir que el tub \u00e9s positiu.<\/span><\/p>\n<p><a href=\"http:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2017\/05\/IMG_3520.jpg\"><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-large wp-image-4593\" src=\"http:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2017\/05\/IMG_3520-1024x768.jpg\" alt=\"IMG_3520\" width=\"1024\" height=\"768\" srcset=\"https:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2017\/05\/IMG_3520-1024x768.jpg 1024w, https:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2017\/05\/IMG_3520-300x225.jpg 300w, https:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2017\/05\/IMG_3520-768x576.jpg 768w, https:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2017\/05\/IMG_3520-200x150.jpg 200w, https:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2017\/05\/IMG_3520.jpg 2016w\" sizes=\"auto, (max-width: 1024px) 100vw, 1024px\" \/><\/a><\/p>\n<p><span style=\"font-weight: 400;\">Aquesta prova, al nostre grup, alguns tubs no ens van acabar de sortir b\u00e9 ja que no vam realitzar la capa d&#8217;anaerobiosi correctament.
