{"id":2814,"date":"2016-05-16T17:41:35","date_gmt":"2016-05-16T16:41:35","guid":{"rendered":"http:\/\/agora.xtec.cat\/ies-merce-rodoreda\/?p=2814"},"modified":"2016-05-17T22:30:40","modified_gmt":"2016-05-17T21:30:40","slug":"control-de-medis-de-cultiu","status":"publish","type":"post","link":"https:\/\/agora.xtec.cat\/ies-merce-rodoreda\/modul-a\/control-de-medis-de-cultiu\/","title":{"rendered":"Control de medis de cultiu"},"content":{"rendered":"<p style=\"text-align: justify;\"><img loading=\"lazy\" decoding=\"async\" class=\" size-full wp-image-2815 aligncenter\" src=\"http:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2016\/05\/img19.jpg\" alt=\"img19\" width=\"600\" height=\"450\" srcset=\"https:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2016\/05\/img19.jpg 600w, https:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2016\/05\/img19-300x225.jpg 300w, https:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2016\/05\/img19-200x150.jpg 200w\" sizes=\"auto, (max-width: 600px) 100vw, 600px\" \/><\/p>\n<p style=\"text-align: justify;\">El control de medis \u00e9s una part important de les activitats que un alumne del cicle formatiu realitza a la formaci\u00f3 professional a l\u2019empresa \u00a0<a href=\"http:\/\/www.aiguesdebarcelona.cat\/ca\/web\/web-aguas-de-barcelona\/inicio\">Aig\u00fces de Barcelona<\/a>.<\/p>\n<p style=\"text-align: justify;\">Els an\u00e0lisis que es realitzen a Aig\u00fces de Barcelona s\u2019utilitzen per determinar la qualitat de l\u2019aigua de diversos punts de control dels rius, l\u2019aigua d\u2019EDARs, ETAPs, o d\u2019altres laboratoris petits que no tenen les instal\u00b7lacions necess\u00e0ries com per a realitzar els an\u00e0lisis necessaris per determinar la qualitat de la seva aigua.<\/p>\n<p style=\"text-align: justify;\"><!--more--><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Tamb\u00e9 es realitzen an\u00e0lisis interlaborals, que s\u00f3n realitzats per diversos laboratoris amb els quals es mira el nivell d\u2019aquests. <\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Aig\u00fces de Barcelona garanteix la qualitat dels resultats dels \u00e0nalisis que obt\u00e9, i \u00e9s per aix\u00f2 que s\u2019ha d\u2019assegurar que tot el material, reactius i medis de cultiu utilitzats estiguin en les condicions adequades; ja que si no, pot ser que els resultats obtinguts no siguin els correctes per culpa del mal estat dels materials.<\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">\u00c9s per aix\u00f2 que s\u2019han de realitzar controls dels medis de cultiu.\u00a0A continuaci\u00f3 explicar\u00e9 en qu\u00e8 consisteix i quina relaci\u00f3 t\u00e9 amb el Cicle de Qu\u00edmica Ambiental.<\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Un medi \u00e9s la base nutritiva que s\u2019utilitza per a fer cr\u00e9ixer els bacteris, i dependr\u00e0 del tipus de bacteri que es vulgui fer cr\u00e9ixer.<\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Existeixen 4 tipus de medis: <\/span><\/p>\n<ul style=\"text-align: justify;\">\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\"><span style=\"text-decoration: underline;\">No selectius:<\/span> contenen suficients nutrients com per soportar el creixement d\u2019una gran varietat de microorganismes.<\/span><\/li>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\"><span style=\"text-decoration: underline;\">Selectius:<\/span> permeten el creixement d\u2019un sol tipus de microorganisme. <\/span><\/li>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\"><span style=\"text-decoration: underline;\">Enriquits:<\/span> S\u00f3n medis no selectius que se\u2019ls agrega subst\u00e0ncies com sang, suero, alb\u00famina, etc. per microorganismes exigents. <\/span><\/li>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\"><span style=\"text-decoration: underline;\">Diferencials:<\/span> s\u00f3n medis de cultiu que permeten establir difer\u00e8ncies entre diferents tipus de microorganismes.<\/span><\/li>\n<\/ul>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Segons el tipus de medi de cultiu s\u2019haur\u00e0 de fer un control o un altre, aix\u00ed com diversos controls per determinar tots els par\u00e0metres dels medis. Alguns dels controls s\u00f3n el control de Productivitat, el control de Selectivitat, o el control d&#8217;Especificitat.<\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">En aquest cas parlarem del Control de Productivitat, que \u00e9s un control que han de passar tots els medis.<\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">El control de Productivitat s\u2019utilitza per determinar si el medi de cultiu permet cr\u00e8ixer el nombre de bacteris correcte, o si en creixen menys degut a les males condicions d\u2019aquest. <\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Per determinar-ho, es compara el creixement de col\u00f2nies de bacteris en el medi sospit\u00f3s, amb un altre medi de comparaci\u00f3. <\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">El Control de Productivitat es realitza durant tres fases diferents, cadascuna en un dia: <\/span><\/p>\n<ul style=\"text-align: justify;\">\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">Reconstituci\u00f3 d\u2019una soca de bacteris (dia 1).<\/span><\/li>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">Banc de dilucions i filtraci\u00f3 (dia 2). <\/span><\/li>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">Recompte de col\u00f2nies (dia 3). <\/span><\/li>\n<\/ul>\n<ol style=\"text-align: justify;\">\n<li style=\"font-weight: 400;\"><strong>Reconstituci\u00f3 d\u2019una soca de bacteris.<\/strong><\/li>\n<\/ol>\n<p style=\"text-align: justify;\"><a href=\"http:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2016\/05\/img21.jpg\"><img loading=\"lazy\" decoding=\"async\" class=\"  wp-image-2816 aligncenter\" src=\"http:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2016\/05\/img21.jpg\" alt=\"img21\" width=\"323\" height=\"430\" srcset=\"https:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2016\/05\/img21.jpg 600w, https:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2016\/05\/img21-225x300.jpg 225w\" sizes=\"auto, (max-width: 323px) 100vw, 323px\" \/><\/a><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Sembrem el bacteri necessari per realitzar el control, i p<\/span><span style=\"font-weight: 400;\">osem la placa a incubar durant un dia. <\/span><\/p>\n<ol style=\"text-align: justify;\" start=\"2\">\n<li><strong> Banc de dilucions i filtraci\u00f3.<\/strong><\/li>\n<\/ol>\n<p style=\"text-align: justify;\"><a href=\"http:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2016\/05\/img17.jpg\"><img loading=\"lazy\" decoding=\"async\" class=\"  wp-image-2817 aligncenter\" src=\"http:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2016\/05\/img17.jpg\" alt=\"img17\" width=\"363\" height=\"272\" srcset=\"https:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2016\/05\/img17.jpg 600w, https:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2016\/05\/img17-300x225.jpg 300w, https:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2016\/05\/img17-200x150.jpg 200w\" sizes=\"auto, (max-width: 363px) 100vw, 363px\" \/><\/a><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Per a dur a terme el control, necessitem diluir la mostra, ja que si no ens sortirien masses col\u00f2nies de bacteris i ens resultaria impossible de contar. <\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Per aix\u00f2, realitzarem un banc de dilucions a partir d&#8217;una col\u00f2nia de bacteris fins a obtenir una diluci\u00f3 10-5.<\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">D\u2019aquest \u00faltim tub agafarem 400 mL repartits en 4 tubs (100mL a cadasc\u00fa). Amb aquests 4 tubs realitzarem la sembra de bacteris als medis.<\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Dos tubs els utilitzarem pel medi a verificar, i els altres dos pel medi de comparaci\u00f3. <\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Filtrem els tubs sobre una membrana (o filtre), i aquest filtre el col\u00b7locarem a sobre del medi nutritiu. Aix\u00ed ens assegurem que tots els bacteris presents en el tub, creixeran sobre la membrana. <\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Posem les plaques a incubar durant un dia. <\/span><\/p>\n<p style=\"text-align: justify;\"><strong>3- Recompte de col\u00f2nies. <\/strong><\/p>\n<p style=\"text-align: justify;\"><a href=\"http:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2016\/05\/img23.jpg\"><img loading=\"lazy\" decoding=\"async\" class=\"  wp-image-2818 aligncenter\" src=\"http:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2016\/05\/img23.jpg\" alt=\"img23\" width=\"340\" height=\"453\" srcset=\"https:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2016\/05\/img23.jpg 600w, https:\/\/agora.xtec.cat\/ies-merce-rodoreda\/wp-content\/uploads\/usu644\/2016\/05\/img23-225x300.jpg 225w\" sizes=\"auto, (max-width: 340px) 100vw, 340px\" \/><\/a><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Agafem les plaques de l\u2019estufa i contem el nombre de col\u00f2nies que han sortit a cada placa. Amb aix\u00f2 podrem comparar el medi a verificar amb el de comparaci\u00f3. Ja que si no es comprova la qualitat del medi i els resultats obtinguts en els an\u00e0lisis no s\u00f3n els correctes, s\u2019hauran de repetir tots. <\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">A les pr\u00e0ctiques de microbiologia del Cicle de Qu\u00edmica Ambiental, no es realitzen controls de medi com a tals, per\u00f2 es preparen medis nutritius, es realitzen sembres de bacteris i filtracions amb membrana. Per aix\u00f2 a mi les pr\u00e0ctiques m\u2019han resultat molt \u00fatils i interessants, ja que he pogut veure una aplicaci\u00f3 laboral de les pr\u00e0ctiques que es fan a l\u2019institut.
